Multi-Enzyme Nanoplatform Offers New Hope for Precision Cancer Therapy

A collaborative research team led by CAS Member Zhang Yaping at KIZ has developed an innovative multi-enzyme-mimicking nanoplatform (MOF-818) capable of inducing multiple programmed cell death pathways simultaneously for enhanced tumor treatment.

Key Innovations:

- Multi-enzyme activity: The nanomaterial mimics peroxidase, catalase, superoxide dismutase, glutathione oxidase, and glucose oxidase activities

- Novel tyrosinase activity: First discovery of tyrosinase-like activity in nanozymes, enabling activation of the common drug acetaminophen (APAP) into potent cytotoxic compounds

- Triple cell death mechanism: Simultaneously triggers apoptosis, disulfidptosis, and cuproptosis

- Targeted delivery: Engineered 3MCT nanoparticles achieve precise tumor targeting through TRAIL receptor recognition

Therapeutic Results:

In animal studies, the combination of 3MCT nanoparticles with APAP significantly inhibited tumor growth and prolonged survival in tumor-bearing mice, with excellent biosafety profiles showing no damage to major organs.
